Reasearch Fellowship (BI) m/f -BIOPOLIS 2024-49 WOLFNESS Job description: Reference: BIOPOLIS 2024-49 Job: Research fellowship (BI) Main Scientific Area: Biological Sciences Associação BIOPOLIS (CIBIO) is opening a call for one Research Fellowship in the framework of the project Preserving the natural heritage of wolves: a multidisciplinary approach towards effective and socially acceptable management of wolf-dog hybridization across Europe (WOLFNESS; DivProtect/0012/2021), funded by FCT through the EU BioDiversa+ Programme. The fellowship is framed in the field of molecular and evolutionary biology, and specifically aims to optimize an amplicon sequencing protocol for genotyping a panel of Single Nucleotide Polymorphisms (SNPs) available for canids and applicable to non-invasive DNA samples. This panel will then be used to assess the prevalence and dynamics of wolf-dog hybridization in the Iberian Peninsula. The selected candidate will carry out wet lab work, including DNA extraction from non-invasive samples and library preparation for Illumina sequencing. Additionally, the candidate will implement bioinformatics pipelines for primer design, PCR multiplexing, and raw data analysis from high-throughput sequencing. 1. Admission requirements and competencies: Candidates should hold a MSc degree in Evolutionary Biology, must be attending an advanced course program in the area of Biological Sciences, and should prove relevant experience in laboratory work related to non-invasive genetics. We seek candidates with i) experience in working with non-invasive samples; ii) skills in genetic data analysis; iii) good oral and written communication skills both in English and in Portuguese; and iv) highly motivated and having teamwork skills. The candidate’s Curriculum Vitae must be compatible with the work plan. It will be also valued presentations at conferences in Evolutionary Biology and international experience. 2. Working plan: The researcher will join the WOLFNESS team, both national and international. The following tasks will be conducted: i) DNA extraction and library preparation of non-invasive samples for Illumina sequencing; ii) primer design and optimization of PCR multiplexes for SNP genotyping; iii) bioinformatic analysis of raw data from high throughput sequencing; 4) assessment of the prevalence and dynamics of wolf-dog hybridization using the SNP data generated; and iv) report writing. The work will be developed under the scientific supervision of Raquel Godinho (PI in BIOPOLIS) and in close collaboration with the WOLFNESS team. 3. Applicable legislation: Law nº.40/2004, 18th August (Research Fellow Statute) amended and republished by Decreto-Lei n.º 123/2019, 28th August; FCT Regulation for Research Studentships and Fellowships ( https://www.fct.pt/apoios/bolsas/regulamento.phtml.pt ). 4. Workplace: The scholarship fellow will be based at the headquarters of Associação BIOPOLIS, CIBIO, Campus de Vairão, Rua Padre Armando Quintas, 7, Vairão, Portugal, and may involve travel in Portugal and abroad in service. 5. Duration of the fellowship: The fellowship will have a duration of 6 months, potentially renewable for additional period. 6. Monthly stipend: Monthly stipend is €1.259,64 according to the stipends established by FCT, I.P. in Portugal ( https://www.fct.pt/wp-content/uploads/2024/02/Tabela-de-Valores-SMM_atualizacao-2024.pdf ). Payment will be made by bank transfer on a monthly basis. The successful candidate will also be entitled to the reimbursement of Social Security payments (Seguro Social Voluntário) and accident insurance. 7. Selection methods: The ranking of candidates will be assessed in accordance with admission requirements and will be based on: a) documental appreciation of the Curriculum Vitae (40%); b) previous experience of the candidate relevant to the project (50%), and motivation letter (10%). The candidate's final classification system shall be given based on a scale of 0 to 100. The jury may interview a short list of candidates to establish the final ranking if the difference between the classification of the top two candidates is less than 10 points. The jury may not award the fellowship if the profile of the candidates does not meet the requirements of the project. 8. A bolsa insere-a na área da biologia evolutiva e molecular, cujo objetivo visa otimizar um protocolo de amplificação e sequenciação para genotipagem de um painel de polimorfismos de nucleótido único (SNPs) disponível para canídeos e aplicável a amostras não invasivas. Este painel de SNPs será depois utilizado para estimar a prevalência e dinâmicas de hibridação lobo-cão na Península Ibérica. O candidato selecionado realizará trabalho laboratorial, incluindo extração de DNA de amostras não invasivas e preparação de biblioteca para sequenciação em plataforma de alto rendimento. Além disso, o candidato implementará análises bioinformáticas para o desenho de primers , multiplexes de amplificação, e análise de dados de sequenciação. 1. Requisitos de admissão e competências : Os candidatos devem ter mestrado em Biologia Evolutiva, estar inscritos num curso avançado na área de Ciências Biológicas, e comprovar experiência relevante em trabalho laboratorial. Plano de trabalho : O candidato vai juntar-se à equipa do WOLFNESS, tanto nacional como internacional. As seguintes tarefas serão desenvolvidas: i) extração de DNA e preparação de bibliotecas de amostras não invasivas para sequenciação de alto rendimento; ii) desenho de primers e otimização de multiplexes de amplificação para genotipagem de SNPs; iii) análise bioinformática de dados de sequenciação de alto rendimento; 4) estimativa da prevalência e dinâmica da hibridização lobo-cão usando os dados gerados; e iv) escrita de relatórios.
